Intracellular NLR (Nucleotide‐binding domain and Leucine‐rich Repeat‐containing) receptors are sensitive monitors that detect pathogen invasion of both plant and animal cells. NLRs confer recognition of diverse molecules associated with pathogen invasion. NLRs must exhibit strict intramolecular controls to avoid harmful ectopic activation in the absence of pathogens. Recent discoveries have elucidated the assembly and structure of oligomeric NLR signalling complexes in animals, and provided insights into how these complexes act as scaffolds for signal transduction. In plants, recent advances have provided (...) novel insights into signalling‐competent NLRs, and into the myriad strategies that diverse plant NLRs use to recognise pathogens. Here, we review recent insights into the NLR biology of both animals and plants. By assessing commonalities and differences between kingdoms, we are able to develop a more complete understanding of NLR function. (shrink)

In billion years of evolution, eukaryotes preserved the chromosome ends with arrays of guanine repeats surrounded by thymines and adenines, which can form stacks of four‐stranded planar structure known as G‐quadruplex (G4). The rationale behind the evolutionary conservation of the G4 structure at the telomere remained elusive. Our recent study has shed light on this matter by revealing that telomere G4 undergoes oscillation between at least two distinct folded conformations. Additionally, tumor suppressor BRCA2 exhibits a unique mode of interaction with (...) telomere G4. To elaborate, BRCA2 directly interacts with G‐triplex (G3)‐derived intermediates that form during the interconversion of the two different G4 states. In doing so, BRCA2 remodels the G4, facilitating the restart of stalled replication forks. In this review, we succinctly summarize the findings regarding the dynamicity of telomeric G4, emphasize its importance in maintaining telomere replication homeostasis, and the physiological consequences of losing G4 dynamicity at the telomere. It seems very commonly agreed that Sappho's wedding-songs display none of the ritual obscenity so frequent in the genre. Thus D. L. Page wrote of fr. i ioa that ‘There is no trace here or elsewhere in Sappho of that ribaldry which was characteristic of the songs recited at this and other stages of Greek wedding-ceremonies’. Similarly Sir Maurice Bowra asserted of fr. 111 that it is ‘neither bawdy nor exalted, but playful. If the humour is a bit primitive, that (...) is due to tradition, which expected jokes at this level’. On the next page he writes of the songs sung outside the bridal chamber, as distinct from those sung in the procession thither, that ‘Here too Sappho seems to avoid bawdry but is not averse from rather elementary jokes’. When I was first invited by Prof. Kim Yong -pyo, editor of the IJBTC, to review this book, I declined, due to the fact that Prof. Park was my teacher and mentor at SUNY Stony Brook, not only as a graduate student, but as an undergraduate as well. For this reason I was afraid that I would not be able to bring the requisite critical distance to the task. After having had the opportunity to read the book, however, I changed (...) my mind, for two main reasons: I realized that it might be personally satisfying to take the opportunity to re-engage myself in the kind of Buddhist soteriological discourse that originally attracted me to Buddhist Studies to begin with, and the potential for lapses in proper critical distance notwithstanding, I felt that there is some sense in which I could bring some insights into the appraisal of this book probably only accessible to myself, given my long relationship with Sung Bae Park and my deep personal interest in his project. So I hope readers of this review will accept it with these factors in mind. There is one question that we may want to ask before dealing with a book like this: Is there a place in our Buddhist Studies academia for the discussion of personal religious experience, or for the investigation of the phenomenon of religiosity? I know of more than a few who would answer such a question with an outright "no." Others might say, "It depends upon how one goes about it." And still others may be very excited by such a prospect. In lieu of an abstract, here is a brief excerpt of the content:Mystery on the Move:Aquinas’s Theological Method as Transforming WisdomGilles Mongeau, S.J.CONTEMPORARY APPROACHES to the thought of Thomas Aquinas have begun to recover its character as a “wisdom practice” aimed at the transformation of persons and sociocultural situations.1 The wise person helps others move along a path through the mysteries of faith toward a wisely ordered life for themselves in a justly ordered society. The starting point of this essay (...) is the supposition that Aquinas’s theological method in the Summa theologiae, rooted in the practice of the three arts of language (grammar, dialectic, and rhetoric) is itself a wisdom practice that proposes an ordered series of exercises that must be performed by the student, leading him or her to a greater participation in divine wisdom. I will show that Aquinas’s use of the tools of rhetoric, particularly the enthymemes known as arguments of fittingness, are a set of practical instructions that facilitate an affective response to divine mystery in order to enable a judgment by connaturality. [End Page 285] Thus, we can learn from Aquinas how fruitfully to order affect and reason in the exercise of the theological craft.The essay proceeds in three parts. First, a thought experiment will help us understand (a) how a medieval text works as a guide to performance, and (b) how intellect and affect interact in both artistic performance and aesthetic appreciation. Second, I will trace the role of affect and reason in knowledge for Aquinas, specifically in knowledge of the beautiful, of the good, and of divine mysteries. While first-person methods are essential for a science of consciousness, it is controversial what form these methods should take and whether any such methods are reliable. I propose that first-person experiments are a reliable method for investigating conscious experience. I outline the history of these methods and describe their characteristics. In particular, a first-person experiment is an intervention on a subject's experience in which independent variables are manipulated, extraneous variables are held fixed, and in which the subject makes a phenomenal (...) judgement about the target experience of the investigation. I examine historical and contemporary examples of first-person experiments: Mariotte’s demonstration of the visual blind spot, Kanizsa’s subjective contours, the Tse Illusion, and investigations of the non-uniform resolution of the visual field. I discuss the role that phenomenal contrast plays in these methods, and how they overcome typical introspective errors. I argue that their intersubjective repeatability is an important factor in their scientific status, however, it is not the only factor. That they control for extraneous factors and confounds is another factor which sets them apart from pseudoscience (e.g., the perception of auras), and hence another reason for classifying them as genuine experiments. Furthermore, by systematically mapping out the structure of visual experience, these methods make scientific progress. Praises of such first-person experimental approaches may not always be sung by philosophers and psychologists, but they continue to flourish as respectable scientific methods nevertheless. (shrink)

Classical temples in ancient Greece show two deterministic illusionistic principles of architecture, which govern their functional design: geometric proportionalism and a set of illusion-strengthening rules in the proportionalism's stochastic margin. Animal morphology, in its mechanistic-deductive revival, applies just one architectural principle, which is not always satisfactory. Whether a Greek Classical situation occurs in the architecture of living structure is to be investigated by extreme testing with deductive methods.Three deductive methods for explanation of living structure in animal morphology are proposed: the (...) parts, the compromise, and the transformation deduction. The methods are based upon the systems concept for an organism, the flow chart for a functionalistic picture, and the network chart for a structuralistic picture, whereas the optimal design serves as the architectural principle for living structure. These methods show clearly the high explanatory power of deductive methods in morphology, but they also make one open end most explicit: neutral issues do exist. (shrink)

In the article "The Epistemological Nature of Information Modeling" [Gnoseologicheskaia priroda informatsionnogo modelirovaniia], Voprosy filosofii, 1963, No. 10, Academician V. M. Glushkov examines two important methodological aspects of the modeling of natural objects and phenomena in the contemporary natural sciences. The first consists of the informational nature of the models created, and the second of their dynamism. The dynamic nature of today's models in the natural sciences means that it is not so much the material structure of the object that (...) is modeled as its behavior and functions in some sphere of natural phenomena. In this sense, the models under examination may also be termed functional, as distinct from the structural models pertaining to a static description of objects. (shrink)

This article presents evidence supporting the claim that ethical reasoning is a skill that can be taught and assessed. We propose a working definition of ethical reasoning as 1) the ability to identify, analyze, and weigh moral aspects of a particular situation, and 2) to make decisions that are informed and warranted by the moral investigation. The evidence consists of a description of an ethical reasoning education program—Ethical Reasoning in Action —designed to increase ethical reasoning skills in a variety of (...) situations and areas of life. ERiA is housed at a public, major comprehensive U.S. university—James Madison University—and assessment of the program focuses on interventions delivered prior to and during orientation for incoming first-year students. Findings indicate that the interventions measurably enhance the ability of undergraduate students to reason ethically. ERiA’s competency-targeted program and positive student learning outcomes offers a promising model for higher education ethics programs seeking to connect classroom learning in ethics to decision-making in everyday life. Fado, the urban folk of Lisbon and Coimbra, is an art of nuances. These nuances music takes from poetry; as ‘sung poetry’ (‘poema cantado’ in Portuguese) fados are not to be equated with ‘songs’ that turn the word into a vehicle – a dominant procedure in, e.g., rock music. Again, ‘voice’ in fado does not so much manifest individual expression; rather it is, as it were, ‘on loan’ from tradition. Keeping some distance from dance, too, fado at the beginning (...) of the 21st century could and can resist being swallowed up into the mainstream of global popular music. (shrink)

In recent years, “deliberation” has become the byword of many political theorists, most of whom identify deliberation with reasoned conversation. Among the most forceful advocates of deliberation as conversation are Jürgen Habermas and, to a greater or lesser extent, his successors who style themselves “deliberative democrats.” For them, the more political decision‐making approximates the ideal of a reasoned public conversation among free and equal individuals, the more legitimate and rational it will be. “Outcomes,” they say are democratically legitimate if and (...) only if they could be the object of a free and reasoned agreement among equals. Their deliberative model produces more rational decision‐making, they say, because it conveys information, impels individuals to order their preferences coherently, and by making persons articulate good reasons in public leads them “to think from the standpoint of all involved.”. Given Conciliar Christology and a compositionalist metaphysics of the incarnation, I explore whether ‘qua-propositions’ are capable of solving the coherence problem in Christology. I do this by probing the metaphysical aspect of qua-propositions, since ‘semantics presupposes metaphysics’. My proposal focuses on the fact that the Word accidentally owns an individual human nature. Due to that individuality, the human properties first characterise the individual human nature and, in a ‘next step’, this individual human nature characterises the Word. I call this ‘stepped (...) characterisation’. Subsequently, I show that stepped characterisation validates the use of qua-propositions in Conciliar Christology. Hence, qua-propositions are not merely ‘muddling the waters of logic’. (shrink)
